Было предложено оценить консервативность аминокислотных остатков в зоне контакта с функциональным лигандом, для чего была сделана выборка из 10 последовательностей ортологов белка SODC_ECOLI. Ниже предствавлена матрица попарного сходства данных последовательностей.

В позициях 117, 126, 135, 138 выравнивания находятся а.о. которые у белка SODC_ECOLI входят в реакционный центр(гистидины 61, 69, 78, аспарагин 81 на рисунке). Помимо указанных остатков есть еще два остака, консервативные не на 100%, но такой уровень консервативности связан с тем, что в 2х последовательностях отсутствовали соответствующие участки последовательности, в остальных а.о. совпали. В выравнивании они занимают позиции 92, 94 (гистидины 44, 46).
Ниже представлено изображение зоны контакта белка с лигандами - ионами меди и цинка. Красным обозначены а.о., консервативные на 100%, синим - более чем на 80%. Видно, что все а.о., взаимодействющие с лигандами, высоко консервативны.
